Mapping of Transposon Insertion Mutants in the Candida albicans Genome

  • Large scale analysis of C. albicans gene function

    The major specific aim of this project is to construct a set of mutant strains of Candida albicans that will be screened by the PI and collaborating laboratories for phenotype alterations and more in depth analysis of specific mutants. A crucial requirement of the technology for construction of the mutant strain is DNA from the site of the targeted mutation that contains the mutation cassette. These DNA constructs and a database supporting them will be provided by TIGR.
  • Generation of C. albicans mutant strains by random transposon insertion

    A 10-12 Kb C. albicans genomic shotgun library is constructed at TIGR. This library is transposed with a transposon (Tn7) containing the mutagenesis cassette (see reference). Clones from the library are sequenced from one direction to establish the site of the transposon insertion. These clones will be provided to generate a collection of mutant strains. By investigating genotype and phenotype correlations, we could explore the genetic basis of fungal virulence and related C. albicans biological traits.
  • Map the genome position of transposon insertion mutants

    To map the insertion sites, we download C. albicans assembly 19 genomic sequences from the Stanford C. albicans sequencing project web site and search these sequences against insertion flanking sequences generated at TIGR. Using a medium stringency cutoff of greater than 85% identity over a minimum 85% insertion flanking sequence length, plus a minimum 300 base pair insertion flanking sequence length, we have tabulated the results of the search. The chromosome location, locus, insertion clone name, flanking sequence accession number, genomic assembly accession number, and the alignment of the insertion flanking sequence with the genomic sequence can be obtained below. We also mapped insertion sites to probable genes annotated by the Canada National Resaerch Council Candida Genome Project (Annotation dataset released on 08/22/2005).
