Total DNA was isolated from the Landsberg erecta accession of Arabidopsis
thaliana. The DNA was sheared and fragments of 0.5-0.7 were ligated into a pUC19
derivitive. A second library, with an average insert of 0.9-1 kbp, was constructed as
well.
Automated sequencing was performed using Big Dye terminators with forward and
reverse primers. Individual bases were assigned phred quality values and the sequences
were assembled using the CAP3 assembly
program. All sequences were trimmed for high quality regions using phred quality
values. Only high quality regions, as defined by three consecutive bases with an average
phred value greater than 30, were parsed out to form high quality contigs.
The current database has been filtered for mitochondrial and chloroplast sequences which
were present in our total Ler DNA preparations. A total of 4,300 high quality contigs
are present in this release.
Please note that we have not performed experimental validation of these polymorphisms
and any candidate polymorphism should be validated by the user.
